연구 중인 비만 치료제 Mazdutide, Innovent Biologics, Inc.

최근 비만 치료제의 강풍이 계속되고 있다.

 

San Francisco와 Suzhou, China를 기반으로 하고 있는 Innovent Biologics, Inc. (Innovent) (HKEX: 01801)의 Mazdutide가 긍정적인 연구 결과를 6월 24일 발표했다. 

 

Mazdutide는 Innovent와 Eli Lilly의 공동 개발되었고, 현재 중국 이외의 해당 약제의 개발에 대한 권리를 Lilly가 소유하고 있다. 

 

Mazdutide는 glucagon-like peptide-1 receptor (GLP-1R)와 glucagon receptor (GCGR)를 동시에 활성화시키는 기전을 가지고 있는 작용제이다. GLP-1R을 활성화시켜 식욕을 감소시키고 위 배출을 지연시켜 체중 감소를 야기할 뿐만 아니라 동시에 GCGR의 활성화를 통해 에너지 소비를 증가시키고 지방산 산화 및 지방 분해를 촉진하며 간 지방을 감소시킨다. 

 

중국인 비만 환자를 대상으로 Mazdutide의 유효성/안전성을 확인한 첫 임상 3상 시험의 연구 (GLORY-1, NCT05607680) 결과가 ADA (American Diabetes Assocation) Scientific session 2024에서 발표되었다. 

투여 후 32주, 48주에 Mazdutide 4mg 및 6mg 투여군이 Placebo군 대비하여 체중 감소 효과를 보였다. 이외에도 심혈관대사관련 위험 요소들이 감소하였고 지방간 수치 역시 감소하였다. 

 

이외에도 여러가지의 임상 3상이 진행 중인데 과체중 및 비만 환자를 대상으로 진행하는 GLORY-1 및 GLORY-2연구, 제2형 당뇨병 환자를 대상으로 진행 중인 DREAMS-1, DREAMS-2 및 DREAMS-3 연구가 있으며 그 중 GLORY-1과 DREAMS-2의 연구는 1차 및 모든 2차 평가 변수를 만족하였다. 

 

DREAMS-1 연구의 결과 발표는 2024년 중반으로 예상하고 있으며, semaglutide와 직접 비교를 하는 연구인 DREAMS-3 연구 결과도 기대되는 바이다.

Innovent는 2024년 2월 중국 NMPA (중국 국가약품감독관리국, National Medical Products Administration)의 CDE(Center of Drug Evaluation)에 Mazdutide의 비만 적응증으로 첫 NDA를 제출하였고 2025년 1분기 허가를 예상하고 있다.

Innovent는 이 외에도 다양한 질환 분야에 대한 제품을 판매 혹은 치료제를 개발 중에 있다. 

 

그 중 종양 분야에서 PD-1 (programmed cell death receptor-1)을 타겟으로 하는 Sintilimab (Tyvyt®)은 Innovent와 Eli Lilly가 공동 개발하였고 현재 중국에서 승인되어 사용되고 있다. 2018년 1월 미국 FDA에서도 Investigational New Drug application를 검토를 승인한 바 있다.
